Tirzepatide is a first-in-class synthetic peptide that simultaneously activates both the GIP (glucose-dependent insulinotropic polypeptide) and GLP-1 (glucagon-like peptide-1) receptors. This dual-agonist design enhances β-cell function, insulin sensitivity, and glucagon regulation.
Research and Phase 3 trials have shown tirzepatide to produce greater reductions in HbA1c and body weight than semaglutide. In the SURMOUNT trial, mean weight loss at 72 weeks reached −15% (5 mg), −19.5% (10 mg), and −20.9% (15 mg) versus −3.1% placebo. Mechanisms include reduced appetite, slowed gastric emptying, and adipocyte-directed lipid flux effects.
